What to look for
In chapter 2's acrostic, the pe verse ('all your enemies open their mouths') comes BEFORE the ayin verse — the alphabet runs in an order no schoolchild learns.

A reversed letter sequence: chapters 2, 3, and 4 of Lamentations all place pe before ayin, while chapter 1 uses the familiar order.
Why it matters
Archaeology vindicated the 'mistake': ancient Hebrew abecedaries dug up at sites like Kuntillet Ajrud show pe-before-ayin was a genuine alternative alphabet order in the biblical period. The lament preserves a fossil of how Israel once recited its letters.
How it has been understood
The midrash, not knowing the archaeology, offered its own beauty: pe is the mouth and ayin the eye — the spies' sin put the mouth before the eye, speaking what they had not truly seen, and the lament for the resulting exile disorders the alphabet to match. Both explanations can be treasured: one for history, one for the heart.
